It was a large temple that received a red seal of 100 stones in the temple territory and served as one of the four temples of the Kanto region of the Shingon sect. The honzon is said to be the work of the monk Gyoki, and it is said that it was called "Kawakami Yakushi" under the belief of Mito Mitsukuni because it flowed upstream when Tokugawa Mitsukuni, the lord of Mito Domain, dumped it into the river. It is not clear how this statue of Yakushi Nyorai came to Yajinji, but it attracted many worshippers as one of the 12 Yakushi of Edo. There used to be Hoju-in, Tokujo-in, Shofuku-in, Hoju-in, Shokaku-in, and Ryuko-in as the head of the temple as the head of the temple, but now only Hoju-in, Tokujo-in, and Ryuko-in remain as temples.
